Accumulation of Cytochrome b6f and PSI Is Not Affected by Loss of LTO1.
Wild-type (WT) and lto1 homozygous plants were analyzed for the accumulation of cytochrome b6f subunits (cytochrome b6, cytochrome f, PetC, and PetD), PC, and PSI subunit PsaA via immunoblotting. Heme staining was also performed to analyze the accumulation of holoforms of cytochrome b6 and f. Total protein samples corresponding to 16 μg of chlorophyll were separated on a 12% SDS-acrylamide gel. For an estimation of the protein abundance in the lto1 mutant, dilutions of the wild-type sample were loaded on the gel. Gels were transferred to membranes before heme staining by chemiluminescence or immunodetection.
