ASAH1 regulates intracellular cAMP levels and ACTH responsiveness. H295R WT or ASAH1KD (pretreated with 5 μg/ml tet for 96 h) cells were treated with 50 nm ACTH for 5 min. Intracellular cAMP levels were quantified by ELISA as described in Materials and Methods. cAMP amounts are normalized to total cellular protein content, and data graphed represent the mean ± sem of three experiments, each done in triplicate.
