BALB/c mice were vaccinated by scarification of the rump with rVVF or rVVG and challenged with RSV. On day 10 post-challenge mice were sacrificed and lungs harvested. Cells were pooled from the mice with in each group and DCs isolated. 1 × 104 DCs per well from rVVF- (i) or rVVG- (ii) primed mice were pulsed for one hour with 10μM OVA peptide (323-339) and cocultured with 105 effector CD4 T cells per well for 96 hours in the presence of anti-OX40L (a, b) or anti-PD-L2 (c, d) or the relevant isotype controls. (a, c) IFN-γ and (b, d) IL-5 concentrations were determined by ELISA. The graphs are representative of two independent experiments and show means ± SD.
