We examined the expression of ALP (A, D, G, J), Bmp4 (B, E, H, K), Bmp2 (C, F, I, L), and P-Smad1/5/8 (N) in the apical cranial mesenchyme at E12.5 and E13.5. ALP was detected by histochemistry, Bmp2 and Bmp4 by in situ hybridization, P-smad1/5/8 by immunostaining. Boxed areas are shown in higher magnification in the image to the right. Note reduced expression of Bmp2 and increased expression of Bmp4 in the mesenchymal (ALP-expressing) layer in Wnt1-Cre; Msx1/2cko/cko mutants. Also note increase in P-Smad1.5.8-positive nuclei (N).
