Strains with deletions of or mutations in the PDI family members exhibit varying sensitivities to the reducing agent DTT. The indicated yeast strains were grown in selective medium to mid log phase. Serial dilutions of the cells were spot plated onto medium containing either 0 mM or 5 mM DTT, as indicated, and grown for 2 d at 30°C. The data are representative of several independent trials, and in liquid culture the doubling times for the single deletions in the absence of DTT were essentially identical.
