A. HSFs were treated with 0.1% DMSO (ctrl) or 100 µM cyclic PFT-α for 30 min, or were subjected to siRNA-mediated silencing of p53 (confirmed by immunoblot using anti-p53 and β-actin as a loading control; right panel). Cells were then stimulated to undergo apoptosis by treatment with CCN1 (2 µg/ml) and/or TNFα (10 ng/ml) for 5 hrs (*p<0.05, n = 3). B. Effect of p53 silencing on apoptosis induced by CHX (1 µg/ml) and/or TNFα for 16 hrs was quantified (NS-not significant, n = 3). C. Immunoblot detection of cytochrome c in cytosolic extracts of cells treated for 4 hrs with CCN1 (C) and/or TNFα (T), or BSA (B), with and without silencing of p53. Numbers under the immunoblots are relative cytochrome c signal intensities as determined by densitometry.
