Quantification of CCK, gastrin, CCK1 receptor, and CCK2 receptor mRNA expression in human pancreatic cancer cell lines by real-time RT-PCR. Human cultured pancreatic cancer cell lines AsPC-1, BxPC-3, Capan-1, MIA PaCa2 and PANC-1 were analyzed for the expression of CCK mRNA (A), gastrin mRNA (B), CCK1 receptor mRNA (C) and CCK2 receptor mRNA (D) by real-time RT-PCR. The presence of CCK peptide in BxPC-3 cells was verified by immunocytochemical staining (E1) with an anti-CCK antibody. Cells were reacted with an anti-human CCK antibody followed by a goat anti-rabbit Alexa Fluor 568 labeled secondary antibody. Negative control cells (E2) were stained with secondary antibody alone. Asterisks (*p<0.05, **p<0.001) indicate values significantly different from that in PANC-1 cells, which express all four gene products. N.C. = not called.
