The absorbance at 215 nm was measured during a 30-ml gradient that reached 1.5 m NaCl. C, CCL16-(1–97) was incubated with recombinant MMP-12 in the presence of heparan sulfate (HS), chondroitin sulfate A (CSA), CSB, or CSC in 5-fold excess (w/w) for 16 h at 37 °C. Cleavage assay products were visualized on silver-stained 15% Tris-Tricine gels.
