By directly inhibiting Rhob expression, miR-21 facilitates Akt1-mediated activation of mTORC1, which promotes cyclin D1 translation initiation. This effect of mTORC1 involves phosphorylation of 4E-BP1, which disables its inhibitory binding of eIF-4E and allows assembly of the eIF-4F complex that additionally contains eIF-4G and eIF-4A (22, 24, 25).
