1μg IgG2a control or anti-DEC-205 mAb were injected intradermally into the ears of C57BL/6 mice. Two days later, targeting antibodies were detected in permeabilised lymph nodes cells by anti-rat IgG / APC. (A) CD11b expression and levels of targeting antibody were evaluated in lymph node cells. (B) CD11b⁺ targeting antibody⁺ cells were gated and expression of MHC class II, CD11c, Langerin and CD8α was evaluated. Percentages of Langerin^neg and Langerin⁺ cells among gated cells are shown.
