SD propagation rate in wild-type (WT) and vesicular Zn2+ transporter ZnT-3 knockout (KO) animals in vitro. A and B: effects of Zn2+ chelation on SD propagation rate. Slices were preincubated with 1 of 2 Zn2+ chelators before SD initiation, TPEN (50 μM; A) or CaEDTA (1 mM; B). Both chelators significantly increased SD propagation rates (*P < 0.01; n = 5). C: in slices from ZnT-3 KO animals (filled bars), control SD propagation rates were significantly higher than those observed in WT slices. In addition, Zn2+ chelation with CaEDTA did not further increase SD propagation rates in ZnT-3 KO slices (*P < 0.01, **P < 0.05; n = 5). D: ZnCl2 (100 μM, 10 min) effectively decreased SD propagation rate in both WT and ZnT-3 KO preparation. Experiments were interleaved between WT and ZnT-3 KO (*P < 0.01; n = 5).
