FIGURE 8. — BACE1 mRNA levels are unaffected by Aβ42 treatment in primary neurons. Triplicate wells of mouse primary cortical neurons were cultured for 7 days and treated with 10 μm oligomeric or fibrillar Aβ42 or appropriate vehicle controls for 24 h. Neurons were lysed and mRNA was isolated and analyzed by TaqMan quantitative RT-PCR for levels of BACE1 mRNA, which was normalized to 18 S rRNA and displayed as percentage of vehicle control. No statistically significant difference between BACE1 mRNA levels in Aβ42-treated or control primary neuron cultures was observed.
