Electromobility shift assays with 6His-SoxS. Purified PCR fragments (10 nM final concentration) were incubated in increasing concentrations of SoxS-6His protein (0 to 1,000 nM) for 20 min at 4°C. The fumC promoter has an established mar/soxbox and acts as a positive control, while the gnd promoter has no such sequence and acts as a negative control. The znuCB upstream regions (−229 to +22 [marked znu 251] and −149 to +22 [marked znu 171]) were amplified as detailed in Materials and Methods. Protein-bound DNA is denoted by an arrow.
