Ang II treatment induced persistent EGFR phosphorylation at tyrosine 845 (Y845) but not tyrosine 1173 (Y1173) in AT1R/Cl4 cells by an Src-dependent mechanism. (A) AT1R/Cl4 cells were rendered quiescent and exposed to Ang II (10⁻⁷ M), EGF (30 nM), or vehicle alone for 10 min, 30 min, 3 h, or 6 h. (B) Quiescent AT1R/Cl4 cells were pretreated with the Src kinase inhibitor PP2 (5 μM) or with vehicle alone for 30 min prior to exposure to Ang II (10⁻⁷ M), EGF (30 nM), or vehicle alone for 10 min. Cell lysates were subjected to immunoblotting with antibodies against Y845- or Y1173-phosphorylated EGFR, followed by stripping and reprobing of the blots with an antibody recognizing total EGFR, as indicated.
