Effect of prostaglandin E2 (PGE2) on the release of vascular endothelial growth factor (VEGF) by human fetal lung fibroblasts (HFL-1). HFL-1 cells were cultured until they reached 90% confluence in 12-well tissue culture plates, after which medium was changed to serum-free Dulbecco's modified Eagle's medium (SF-DMEM), with or without PGE2. (A) Concentration dependence. After 2 days, the monolayer culture medium of HFL-1 stimulated by varying concentrations of PGE2 was harvested and assayed for VEGF by ELISA. Vertical axis, release of VEGF (picograms per 105 cells per day). Horizontal axis, concentration of PGE2 (molar). (B) Time course. Monolayer cultures were stimulated with PGE2 (1 × 10−7 M), and the medium was harvested at varying time points and assayed for VEGF by ELISA. Vertical axis, release of VEGF (picograms per 105 cells).
