MOSC2 and CYB5B are essential for the amidoxime reductase activity. A, efficiency of siRNA-mediated down-regulation of MOSC2, CYB5A, CYB5B, CYB5R3, CYB5A + CYB5B, and CYB5B + MOSC2 in differentiated 3T3-L1 adipocytes as determined by Western blot analysis. Cell lysates prepared from siRNA-transfected adipocytes were separated by SDS-PAGE and analyzed by Western blot using specific antibodies as shown. Calnexin is included as a loading control. Quantification of the down-regulated proteins is shown in supplemental Fig. S7. B, effect of siRNA-mediated down-regulation of MOSC2, CYB5A, CYB5B, CYB5R3, CYB5A + CYB5B, and CYB5B + MOSC2 in differentiated 3T3-L1 adipocytes on the benzamidoxime reductase activity.
