Ypk2 physically associates with TORC2. The interaction between Ypk2 and each TORC2 component was determined by the GST pulldown experiment. Lysates were prepared from cells expressing GST or GST-Ypk2 along with one specific epitope-tagged TORC2 component and incubated with glutathione-Sepharose beads. Proteins bound on the beads were separated and detected by Western analysis using anti-HA or anti-Myc antibodies to detect the tagged TORC2 component. GST and GST-Ypk2 in the pulldown samples were visualized by Coomassie Blue staining. The expression levels of the HA- or Myc-tagged TORC2 components in the lysates are shown in the bottom panels.
