SapC interacts with CD1c-Fc in a liposome-proximal topology. (A and B) Saposins and/or CD1 fusion proteins were incubated with liposomes (2 mM) at pH 5 as indicated. Samples were transferred to a sucrose gradient buffered to pH 5 (or pH 7) and centrifuged at 200,000 × g. Gradient fractions were collected, separated by SDS/PAGE, and visualized by silver staining.
