After two weeks in culture, total RNA was extracted from homogenized hESC-seeded scaffolds. Expression of genes representative of the mesoderm (a), endoderm (b) and ectoderm (c) germ layers was measured by RT-PCR and normalized against GAPDH expression levels. Ratios of normalized values relative to their expression levels in suspended EBs were calculated and are presented as the mean values (±SD) of three experiments, each performed in quadruplicates.
