The transcriptional activity of BnAP2 was determined using a yeast assay system in which BnAP2 was expressed in yeast strain AH109 containing a lacZ reporter gene. A, BnAP2 constructs. Six constructs which were fused in frame to the GAL4 DNA binding domain (DB) expression vector were prepared containing various portions of BnAP2. B, Filter lift β-galactosidase assay was performed. C, The liquid β-galactosidase assay was performed using ONPG as a substrate. β-Galactosidase activity was then assayed and expressed in Miller units.
