Med1+/− Med24+/− double haploinsufficiency blocks estrogen-dependent gene expression and estrogen-stimulated mammary duct growth. (A) Gene expression levels. Primary mammary epithelial cells were isolated from wild-type and mutant mice as indicated and treated with 10−7 M E2 for the indicated time. Total RNAs were isolated and expression of the indicated ERα target genes was measured by real-time PCR. Values (means ± SD from a representative experiment performed in triplicate) are plotted as a fold increase against the value obtained before E2 addition. (B) Mammary duct growth. Three-week-old wild-type and mutant mice as indicated were ovariectomized and implanted with slow-release E2 pellets for 21 days. Whole-mount stainings of inguinal glands from these mice are shown. Scale bar, 1 mm.
