Engineered expression plasmid for RC mutagenesis. Segments of the L and M genes targeted for mutagenesis (striped regions and Table 1) are flanked by unique restriction sites that facilitate the insertions of oligonucleotide cassettes encoding multiple substitutions. Fifteen restriction sites (parentheses) in the plasmid were removed to enable the cassette-based mutagenesis approach.
