Comparison of cell numbers determined with qPCR assays and the Thoma-Zeiss counting chamber using human adult feces spiked with Bifidobacterium species cells. (A) Spiking was performed with 6 log10 (▲), 7 log10 (●), and 8 log10 (■) cells of six Bifidobacterium species. (B) Spiking was performed with 5 log10 (n = 3), 6 log10 (n = 6), 7 log10 (n = 6), 8 log10 (n = 6), and 9 log10 (n = 5) cells of B. adolescentis (⊙), B. angulatum (⊡), B. animalis (Inline graphic), B. breve (♢), B. pseudolongum (▽), and B. thermophilum (○). The values are means. Significance was estimated by one-way ANOVA with Bonferroni's posttests. Differences of means were not significant. GroEL copy recovery was 48% ± 6% (A) and 45% ± 6% (B) (means ± SD).
