Coupled enzymatic scheme for assaying the activity of FDPS. FDPS converts one IPP and one DMAPP molecule into one GPP molecule, then converts that GPP molecule into FPP with an additional IPP molecule. Once the FPP is generated, PFTase catalyzes the attachment of farnesyl to a dansylated CAAX peptide. Measuring the fluorescence intensity at an excitation wavelength of 340 nm and an emission wavelength 505 nm allows the direct monitoring of prenylated peptide which is directly related to the amount of FPP produced by FDPS.
