A and B, HEK293 cells were transfected with HA-PAR4-Luc (0–0.1 μg) (A) or V5-PAR4-GFP (0–0.25 μg) (B) and assayed for luciferase activity with colenterazine H (5 μm) or GFP expression to determine the minimal amount of plasmid required for BRET assays. C, HEK293 cells were transfected with HA-PAR4-Luc (0.03 μg) and V5-PAR4-GFP (0–0.24 μg). Expression levels were determined by quantitative flow cytometry with anti-HA or anti-V5 antibodies. HA-PAR4-Luc is plotted against increasing expression of V5-PAR4-GFP.
