NO and superoxide production by wild-type and the ZnS4 cluster mutants of eNOS. Recombinant wild-type eNOS and the ZnS4 cluster mutants (C94A, C99A, and C9499A) were incubated with L-arginine and the appropriate co-factors for 30 min at 37 °C and NO (A) and superoxide levels (B) determined as described (Grobe et al. 2006). There is a significant reduction in both NO (A) and superoxide (B) generation in the ZnS4 cluster mutants. Data are presented as mean±S.E.M.; n=3. *P<0·05 versus wild-type eNOS.
