Deletion of C- and N-terminal regions reduces activity. (A) The AUF1 or AUF1-ΔQRGG (dQRGG) expression plasmids were cotransfected into RAW-264.7 cells with VEGF-3′ UTR-Luc (see Figure 5B). Data are normalized to cells transfected with the empty expression plasmid pcDNA-3.1 (pcDNA). Deletion of the QRGG region inactivated the inhibitory effect of AUF1 on VEGF-3′ UTR-Luc. p < 0.0001, pcDNA vs. AUF1; pcDNA vs. ΔQRGG was not significantly different. p < 0.0001 AUF1 vs. ΔQRGG. (B) Deletion of 21 aa (AUF1-Δ21-Ala, d21) from the polyalanine region of the full-length AUF1 inactivated AUF1.
