Rhodamine 123 accumulation assay. Functional impact of gain-of-function β-catenin was tested by incubating HCT116, HAB-68mut and HAB-92wt cells for 10 min (A) and 15 min (B) with rhodamine 123. Fluorescence intensity of the cells was measured after another 60 min in rhodamine 123-free medium. Despite their different β-catenin genotype and ABCB1 expression, no differences in accumulation of rhodamine 123 were measured.
