IGF-I modulation of CNG channels expressed in Xenopus oocytes requires a specific tyrosine residue. cGMP-sensitivity of expressed CNG channels in excised patches from control and IGF-I-treated (30 min pretreatment with 1 μM IGF-I) oocytes. (A) IGF-I significantly reduced the K1/2 of wild-type bRET channels (P < 0.005; n = 6–8) but did not change the K1/2 of bRET channels in which Y498 was replaced with a phenylalanine (P < 0.05; n = 8–15). (B) IGF-I did not significantly change the K1/2 of wild-type rOLF channels (P < 0.001; n = 4–6) but did reduce the K1/2 of rOLF channels in which F477 was replaced with a tyrosine (P < 0.01; n = 6–12).
