To demonstrate the requirement of TXNIP for d-flow-mediated adhesion molecule expression, the thoracic aorta (s-flow region) and the lesser curvature of the aortic arch (d-flow region) from EC-TXNIP KO mice and control littermates were harvest for RNA extraction. mRNA expression of VCAM-1 (A) and ICAM-1 (B) was detected by quantitative real-time PCR. Data are expressed as mean ± SD from 7 mice in each group. *P<0.05. (C) HUVEC were transfected with either scramble control or TXNIP-specific siRNA for 48 h. Then EC were exposed to d-flow or kept at static condition for an additional 24 h. Protein expression of VCAM-1, ICAM-1 and TXNIP was analyzed by Western blot.
