AFM imaging demonstrates that StpA forms a rigid protein filament along DNA resulting in simultaneous stiffening and bridging of DNA. (A) Air AFM topology imaging of linearized φX174 dsDNA incubated with 1 StpA per 1 bp (1:1 StpA:DNA ratio, 300 nM StpA) shows rigid StpA-coated DNA hairpins. (B) Naked φX174 DNA on the same type of surface. Comparing with A, the naked DNA assume random coiled conformations with much thinner backbones. (C) DNA–StpA complexes at 1:10 StpA:DNA (30 nM StpA) ratio suggesting localize StpA binding and partial DNA condensation.
