Cellular immune responses of HF10-immunized mice to HSV-2 proteins. Splenocytes (1 × 106 cells) from HF10-immunized or unimmunized mice were collected and incubated at 37°C for 3 h. NIH3T3 cells (1 × 106 cells) transfected with expression vectors for HSV-2 gD, ICP0, and UL46 were added into dishes to stimulate splenocytes. To confirm protein synthesis, 186-infected NIH3T3 cells were treated with either 10 μg/ml ganciclovir (186/GCV) or 20 μg/ml cycloheximide (186/CHX) for 8 h and used for stimulation. The media were collected at 5 and 20 h after addition and assayed for IFN-γ concentrations by ELISA. “Cells” denote uninfected NIH3T3 cells used as a negative control.
