ROS production induced by cholesterol was prevented by the inhibition of mitochondrial respiration complex I. A, Mitochondria isolated from Hepa1c1c7 cells were incubated with cholesterol (10 μm) bound to BSA in the presence or absence of rotenone (10 nm) for 5 min as noted. Carboxy-H2-DCF-DA was then added to the mitochondria for 30 min, followed by measurements of ROS as detailed in Materials and Methods.
