PMA enhances recruitment of NF-κB proteins p50 and p65 to the GRK5 promoter. NRVMs were treated with 20 nm PMA and cross-linked with 1% formaldehyde followed by lysis and sonication. Chromatin solutions were immunoprecipitated using antibodies specific for p50 and p65. DNA obtained from chromatin solution, was quantified by real-time PCR using primers indicated by the arrows in A for the relative abundance of a 112-bp region surrounding the NF-κB site on the GRK5 gene (see “Experimental Procedures”).
