p38, JNK MAPK, PI3K-Akt, Src family of tyrosine kinases, and NFκB activation are necessary for ActApo-induced up-regulation of maturation markers on DCs. Day 6 immature DCs were preincubated with either vehicle alone or the different MAPK inhibitors SB203580, U0126, or SP600125 (A), the PI3K inhibitor wortmannin (B), the Src family tyrosine kinase inhibitors PP1 and PP2 (C), and the IKK inhibitor wedelolactone (D) for 30–60 min and then left untreated (Med) or co-cultured with ActApo. Expression of maturation markers was measured by flow cytometry, and data are represented as MFI of CD80 and CD86 expression on multiple DC donors (n = 11 (A), n = 8 (B), n = 9 (C), and n = 8 (D). Significant differences comparing the ActApo with ActApo in the presence of inhibitor were assessed by non-parametric Wilcoxon test and are indicated by ** (p < 0.01) and *** (p < 0.001).
