TPR, catalytic domain deletion mutant; ΔTPR, TPR domain deletion mutant; GST, GST tag only. B, a GST pull-down assay was performed using WT and PP5 deletion mutants. A GST tag was used as a control. These proteins (25 μg) were incubated with glutathione-Sepharose beads and S100A1, S100A2, S100A6, or S100B protein in a presence of 1 mm CaCl₂ or EGTA. After agitation at 25 °C for 1 h, the beads were washed and then eluted with SDS sample buffer. The eluted samples were analyzed by 10% Tricine SDS-PAGE. The gel was stained with CBB.
