T0901317 rapidly reverses the suppressive effect of arachidonate on SREBP-1c mRNA. On day 0, FTO-2B cells were set up in medium A containing 5% FCS. On day 3, the cells were switched to medium B and preincubated for 6 h in the absence or presence of 100 μM sodium arachidonate in a final concentration of 0.1% BSA. Two groups of cells were then harvested (lanes 1 and 2), and the remaining cells (lanes 3–8) were treated with either 1 μM T0901317 in 0.1% DMSO or 0.1% DMSO alone for the indicated time. Total RNA from three dishes was pooled and isolated. Aliquots of total RNA (20 μg) were hybridized for 10 min at 68°C to the indicated ³²P-labeled cRNA probe. RNase-protected fragments were separated by gel electrophoresis and exposed to film at −80°C for 6 h.
