Figure 7

Arachidonate competitively inhibits LXR activation by 24(S),25-epoxycholesterol as measured by fluorescence polarization. (A) The indicated concentration of sodium arachidonate (20:4) was evaluated for its ability to inhibit the binding of an LXXLL-containing peptide to LXRα in the presence of the indicated concentration of 24(S),25-epoxycholesterol. Data are presented as the increase in fluorescence polarization (mP) normalized to the BSA control. (B) Eadie–Hofstee plot of the data in A. A blank value for fluorescence polarization in the absence of 24,25-epoxycholesterol was subtracted from all measured values.
