MSC express iNOS and produce NO in response to lymphocyte stimulation. (A) 1 × 105 ConA-stimulated PVG LNC were cultured in the absence (CTR) or presence of syngeneic PVG.7B MSC [log (MSC:LNC)] and expression of iNOS was analyzed after 24 h. iNOS was detected in co-cultures of ConA–LNC with MSC and correlated with the numbers of MSC present. (B) In the same set-up as shown in (A) the amount of nitrate (shown as the mean and the SD of triplicates) as a proxy for NO concentration was determined in the absence (□) or presence (Inline graphic) of L-NMMA. Addition of L-NMMA abolished NO levels produced in MSC:LNC co-cultures. NO was not detected in the absence of MSC (CTR). Data are representative of three independent experiments.
