Classical DC licensing for cross-priming is mediated by Th cells and enhanced by TLR ligands. DCs interact with Th cells via antigen that is presented on MHC class II and recognized by the TCR and via CD40–CD40L interactions. The signals downstream of CD40 and TLRs lead to the upregulation of MHC class I and CD80/86, thus allowing the DC to interact with CTL.
