MVA-expressed Ag85A is not efficiently presented by ALDC. FACS-sorted FSChigh DEC-205+ cells from MHC-defined animals were infected with recombinant viruses expressing Ag85A or loaded with rAg85A (2 μg/ml) or PPD-B (20 μg/ml) and were used as antigen-presenting cells. IFN-γ responses by PBMC from MHC-matched animals (n = 8) were measured by ELISPOT assay. Bars represent mean numbers of spot-forming units (SFU) per million PBMC.
