Activity assays using purified γ-secretase and C100-FLAG substrate were performed in detergent-free vesicles comprising different ratios of various FA-containing PCs at a total lipid concentration of 1.5 mM. Assay solutions were analyzed by A) AICD-FLAG Western blot densitometry as a measure of total γ-activity, B) Aβ40 ELISA, and C) Aβ42 ELISA,,enabling the calculation of D) Aβ42/40 ratios for the POPC background, and E) for the SOPC background. Ratios given in the key denote the percentage of POPC (first number) and of specific FA being tested (second number). Error bars, standard errors. Data were analyzed by one-way ANOVA with Bonferroni post-test correction. a/b/c (comparing to 100% control lipid) and */**/*** (comparing between lipids) denote statistical significances of p<0.05, p<0.01, and p<0.001, respectively (n = 3-12).
