Synthesis of antibody-decorated nanoparticles. AlexaFluor 647 (AF647)-tagged lipids (1) as well as gadolinium lipids (3) were synthetized. Nanoparticles were then formed with maleimide polyethylene glycol (PEG) lipids (2), PEG lipids, ω-carboxynonanoylcholesteryl ester, and phosphatidylserine (not shown). Nanoparticles (4) were subsequently conjugated with polyclonal anti-myeloid-related protein (Mrp)-14 or nonspecific IgG antibodies via maleimide-sulfhydryl chemistry.
