CGA-Luciferase activation downstream of GnRH requires Nfat4. LβT2 cells were transiently transfected with a scrambled sequence NS siRNA or siRNA specific for Nfat4 and a −1500/+45 CGA-Luciferase reporter construct. Seventy-two hours after transfection, LβT2 cells were treated with vehicle (PBS) or GnRH (10 nm) for 8 h. The cells were lysed, and the luciferase activity was measured. Data were normalized as described by Valcu and Valcu (65). Data shown are the mean ± sem for three independent experiments each performed in triplicate.
