Selective siRNA knockdown of Cx43 and Cx45 expression blunts hypotonicity-induced ATP release from CFs. A) Cx43 siRNA knocked down Cx43 mRNA expression by 91% 24 h after transfection (P<0.001; SCR, scramble siRNA). B) Cx43 protein levels were decreased >90% 24-48 h after transfection with Cx43 siRNA. C) CFs transfected with Cx43 siRNA (assayed 48 h post-transfection) released 47% less ATP than controls after 5 min hypotonic stimulation (P<0.001).
