A. CFTR Cl⁻ current was measured in FRT cells expressing wild type CFTR-expressing in the presence of a transepithelial Cl⁻ gradient and after basolateral membrane permeabilization. CFTR Cl⁻ current was stimulated by forskolin (20 µM), followed by eugenol addition. CFTR Cl⁻ current was completely inhibited by CFTR_inh-172 (10 µM). B. Investigation of possible synergy/competition of eugenol with T16A_inh-A01. T16A_inh-A01 (1 µM) was added to inhibit TMEM16A Cl⁻ current by ∼50% followed by indicated concentrations of eugenol. C.
