Effects of antioxidants, Nox inhibitor, H2O2 scavenger, and AT1R antagonist on Ang II-induced changes in
CYP11B2
mRNA and protein expression in H295R cells. Cells were treated with Ang II (10 nM for 6 h) in the absence and presence of either antioxidant (NAC, 200 μM) or H2O2 scavenger (PEG-Cat, 350 U/ml) or Nox inhibitor (VAS-2870, 20 μM) or losartan (10 μM). Cells not treated with Ang II were used as CON. Cell lysates were analyzed for CYP11B2 mRNA expression by real-time PCR (A), and CYP11B2 protein expression was analyzed by western blot analysis (B). The protein and mRNA expression were normalized using β-actin and 18S, respectively. Results derived from six independent experiments are presented as mean±SEM. ** and ##p<0.01. ** denotes a significant difference between the control and Ang II treated samples. ## denotes a significant difference between Ang II and Ang II+drug-treated samples.
