STAT3 inhibits tumor invasion by suppressing expression of SNAI. A, STAT3 suppresses expression of SNAI in Apcmin/+ mice. Expression of the indicated proteins was measured by immunoblotting from the isolated tumor cells from the small intestines. B, STAT3 suppresses expression of SNAI in human CRC lines. STAT3 was silenced by transfection of siRNA, and expression of the indicated proteins in the indicated CRC lines was measured by immunoblotting 3 days after transfection. C, STAT3 suppresses expression of SNAI in a human CRC cell line. HCT116 cells were transfected with either GFP or GFP-STAT3-WT, and the levels of the indicated proteins were measured by immunoblotting. D, SNAI mediates the invasion of the STAT3-depleted HCT116 cells. Cancer cell invasion assay was performed as described under “Experimental Procedures” 3 days after transfection of the indicated siRNA (S3, STAT3; SN, SNAI).
