PH-truncated 261-p97Eps8 reduces LPS-induced activation of FAK, Src, and p38 MAPK as well as phagocytosis of E. coli and latex beads. A, RAW cells and their derived Ctrl and 261-p97Eps8-expressing (261-9 and 261-10) cells were treated with PBS (−) or LPS (100 ng/ml) for 24 or 48 h. The lysates (100 μg) were resolved by SDS-PAGE and probed with antibodies as indicated. The asterisk indicates the position of active Src. Phagocytosis of GFP-E. coli (B) or latex beads (C) in PBS-pretreated (−) or LPS-pretreated RAW, Ctrl, and 261-9 and -10 cells was done exactly as described above.
