Strains harboring mutations in DIS3, MTR3, and MTR4 accumulate mitotic spindles. (A) Examining microtubule architecture in exosome subunit and cofactor gene mutant cells. Wild-type or mutant cells expressing GFP-TUB1 were fixed and stained with DAPI to visualize the DNA. Arrows, pre-anaphase spindles; arrowheads, anaphase spindles. (B) Quantitative analysis of mitotic spindle accumulation phenotype in mtr mutants. Cells were scored for the presence of an anaphase or preanaphase spindle at either the permissive (23°) or restrictive (37°) temperatures. At the restrictive temperature, these cells arrest growth and accumulate polyA+ RNA in the nucleus (Kadowaki et al. 1994).
